In this study, we sought to develop a reliable, specific and high-throughput method for the quantitative analysis of conformational changes of GLUT-1 molecules. In order to label GLUT-1 molecules, we employed a novel monoclonal antibody that specifically binds to an epitope of GLUT-1, the conformational change of which accompanies glucose transport activity. The monoclonal antibody-based probe was designed to be conjugated with 1,2-dioleoyl-sn-glycero-3-phosphoethanolamine (DOPE), a membrane anchor. In order to test the ability of the probe to quantify the conformational change of GLUT-1 molecules, HEK293 cells expressing GLUT-1 with insulin treatment were incubated with the probe. The cells were stained with secondary antibody conjugated with Cy5 fluorophore and analyzed by flow cytometry. The staining efficiency of the probe was determined by setting up control experiments to calculate the change in mean fluorescence intensity upon insulin treatment. The dye-exclusion assay was conducted to examine the specificity of the probe. The assay for quantitative analysis of conformational change of GLUT-1 molecules showed good linearity (R(2)=0.99) and was able to distinguish a change in fluorescence intensity greater than two-fold from the vehicle.
